Necessary and sufficient for commitment of CD4 lineage, while its absence causes CD8 commitment. Development of immature T-cell precursors (thymocytes) to either the CD4 helper or CD8 killer T-cell lineages correlates precisely with their T-cell receptor specificity for major histocompatibility complex class II or class I molecules, respectively. Immunofluorescence: HeLa cells were fixed with 4% PFA (20 min), permeabilized with Triton X-100 (0.1%, 10 min), then stained with ARG55025 anti-ZBTB7B antibody (green) at 1:25 dilution, 1 hour at 37°C. Cytoplasmic actin was counterstained with Alexa Fluor® 555 (red) conjugated Phalloidin (7 units/ml, 1 hour at 37°C). Nuclei were counterstained with DAPI (blue) (10 µg/ml, 10 min).
